Top 10 Pharmaceutical Excipients Used in Tablet Manufacturing in India

3 February 20258 min read

Whether you are developing a direct compression tablet or a wet granulation formulation, these are the 10 pharmaceutical excipients that form the foundation of virtually every tablet made in India.

India produces approximately 20% of the world's generic medicines by volume, making it the pharmacy of the world. At the heart of every tablet manufactured is a carefully selected blend of pharmaceutical excipients β€” inactive ingredients that determine the physical properties, manufacturing behaviour, stability, and patient acceptability of the final dosage form. Here are the 10 most important excipients for tablet manufacturing, all of which are stocked by Surravi Phharma in Hyderabad.

1. Mannitol β€” The Premier Direct Compression Diluent

Mannitol is the gold-standard diluent for orally disintegrating tablets (ODTs) and chewable tablets due to its non-hygroscopic nature, mild sweetness, cool mouthfeel, and excellent compressibility. At 98.0–102.0% assay, IP/BP/EP/USP grades are available. It is also used in injectable formulations as an osmotic agent. For any formulation requiring a non-cariogenic, diabetic-safe sweetener-diluent combination, Mannitol is the first choice.

2. Lactose Monohydrate β€” The Universal Tablet Filler

Lactose remains the most widely used pharmaceutical diluent globally. It is compatible with most APIs, cost-effective, and available in grades optimised for direct compression (spray-dried) and wet granulation. Its limitations β€” incompatibility with primary amines and unsuitability for lactose-intolerant patients β€” are well-known and easily managed through formulation design.

3. PVP K30 (Povidone K30) β€” The Versatile Binder

PVP K30 is the most commonly used binder in wet granulation, used at 2–5% dissolved in water or isopropyl alcohol. Beyond binding, it enhances dissolution of poorly soluble drugs by forming solid dispersions and inhibiting API recrystallisation. It is compatible with virtually every API and most other excipients, making it the first-choice binder for new tablet formulations.

4. HPMC (Hypromellose) β€” Film Coating and Sustained Release

HPMC serves two distinct roles depending on the viscosity grade. Low-viscosity grades (E5, E15) are film coating polymers used in aqueous coating systems. High-viscosity K-grades (K4M, K15M, K100M) are hydrophilic matrix formers for sustained-release tablets. HPMC is also the base material for vegetarian (HPMC) capsule shells, replacing gelatin in vegan and halal-compliant products.

5. Aerosil (Colloidal Silicon Dioxide) β€” The Essential Glidant

Used at just 0.1–0.5%, Aerosil dramatically improves powder flow by reducing interparticle friction. It is indispensable in direct compression formulations and for APIs with poor flow properties. Its high surface area (175–225 mΒ²/g) also makes it an effective adsorbent for liquid APIs and a thickening agent in topical gels.

6. Citric Acid Anhydrous β€” Effervescent and Buffer Systems

Citric Acid Anhydrous is the primary acidulant in effervescent tablet and sachet formulations, reacting with Sodium Bicarbonate to produce COβ‚‚. It is also used as a pH modifier, chelating agent, and taste enhancer in oral liquids. The anhydrous form is strongly preferred in effervescent manufacturing over the monohydrate to prevent premature acid-base reaction from moisture.

7. Talc β€” Lubricant and Anti-Adherent

Talc is a multi-functional excipient used as a glidant (improving flow), lubricant (reducing tablet-punch friction), and anti-adherent (preventing tablet sticking to punch faces). Used at 1–5% in tablet formulations, it is compatible with virtually all APIs and other excipients. It also acts as a diluent in some formulations.

8. Sucralose β€” High-Intensity Sweetener for Taste Masking

At 600Γ— the sweetness of sucrose, Sucralose is the preferred sweetener for bitter API taste masking in chewable tablets, oral liquids, and dispersible formulations. It is calorie-free, diabetic-safe, pH-stable, and thermally stable β€” properties that make it ideal for both solid and liquid dosage forms. It is used at 0.01–0.05% concentration, adding negligible weight to formulations.

9. Maltodextrin β€” Spray Drying Carrier and Bulking Agent

Maltodextrin is the most widely used carrier in spray drying applications β€” used to convert liquid extracts, APIs, and nutraceutical ingredients into free-flowing powders. In tablet formulations it serves as a binder and diluent. In nutraceutical products it is the standard bulking agent in protein powders and meal replacements.

10. Sodium Bicarbonate β€” Effervescent and Antacid Applications

Sodium Bicarbonate (NaHCO₃) is the alkaline component of effervescent tablet systems, reacting with Citric or Malic Acid to generate COβ‚‚. As an antacid API it neutralises gastric acid, making it both an active ingredient and an excipient depending on the formulation. It is also used as a pH modifier in oral liquid formulations.
