What are Gelatin Capsules?
Gelatin capsules are made from animal-derived gelatin — a protein obtained by partial hydrolysis of collagen from bovine (cattle) or porcine (pig) hides and bones. They have been the industry standard for pharmaceutical hard capsules for over 150 years. Gelatin capsule shells typically contain 13–16% moisture and have a smooth, transparent appearance. They are available in sizes 5 (smallest, ~0.13 mL) through 000 (largest, ~1.37 mL), and in both plain transparent and a wide range of opaque and two-tone colours.
What are HPMC Capsules?
HPMC capsules (also called vegetarian capsules, veggie caps, or hypromellose capsules) are made from Hydroxypropyl Methylcellulose — a semi-synthetic cellulose polymer derived from plant sources. They contain significantly less moisture (3–7%) than gelatin capsules and do not require animal-derived ingredients, making them suitable for vegetarian, vegan, halal, and kosher-certified products. HPMC is listed in the IP, BP, EP, and USP as an approved capsule shell material.

Moisture content is the most practically significant difference: gelatin shells contain 13–16% moisture while HPMC shells contain only 3–7%. This makes HPMC capsules significantly more suitable for moisture-sensitive APIs — hygroscopic drugs, lyophilised powders, and formulations containing moisture-reactive excipients. Gelatin is more susceptible to cross-linking reactions when exposed to aldehydes (from excipients or packaging), which can form an insoluble pellicle that slows capsule dissolution significantly. HPMC does not undergo aldehyde cross-linking.

In India's context, HPMC capsules are increasingly preferred for nutraceutical and Ayurvedic products because a significant proportion of the Indian population is vegetarian. For products targeting Muslim consumers (where porcine gelatin is forbidden under halal dietary law) or Jewish consumers (kosher dietary law), HPMC is the required choice unless bovine halal-certified gelatin is specifically sourced and certified. For pharmaceutical prescription products in India, gelatin capsules are still the dominant format, but HPMC adoption is growing rapidly.

Gelatin capsules have better machinability than HPMC on legacy high-speed capsule filling machines — they have predictable locking geometry and are very consistent dimensionally. HPMC capsules may require machine setting adjustments, particularly at high speeds, and are generally slightly more expensive than equivalent gelatin capsules.
